Repeat of DMJ Valproate and Saccharin CTA and DML Lower Valproate and Saccharin CTA but with 60 mg/kg dose of valproate instead of 200 or 20 mg/kg dose. 60 mg/kg is half log unit between 20 and 200 mg/kg.

Medium Dose Valproate Dilution

15 ml of 0.15M Sodium Valproate + 35 ml of 0.15M NaCl (to maintain 300 mOsm tonicity)

--> 50 ml of 45mM Sodium Valproate (a tenfold dilution), at 2.5 mg/ml.

8 ml/kg = 60 mg/kg

Rats arrive (September 28, 2021) n=16 adult male (8) and female(8) sprague-dawley rats (Charles River CD), individually housed in BRF 169.


Day 0: Water removed ( October 5, 2021)


Days 1 - 8: Water Restriction ( October 6-13 , 2021)

Weigh rats daily. Water for 30 min, ad lib food.

30 min water at 1430 daily.


Day 9: Conditioning Day (October 14, 2021)

-15 min: All rats injected with 15mM Na Valproate 8ml/kg = 20mg/kg 0 min: 30 min access to 0.125% saccharin (5 g sodium saccharin/4L water) 30 min: all rats injected with 0.15 M NaCl, 3 ml/kg ip. 40 min: half of rats injected with 0.15 M LiCl 3mlkg ip; other half of rats 0.15 M NaCl 3 ml/kg ip.

ad lib water returned after final injection overnight.

  • rats 1-8 : female, Rats 9-16: male
  • rats 1-16: Valproate;
  • All rats: saccharin
  • rats 1-4,9-12: 3ml/kg of 0.15M LiCl; rats 5-8,13-16: 3ml/kg of 0.15M NaCl


Days 12 - ???: Preference Tests (October 15 - ??? , 2021)

Rats CTA acquisition and extinction was measured with 24-h, 2-bottle preference tests. Rats were given 24-h free access to both 0.125% saccharin and water bottles, alternating position for 12 days.
