Procedure

Sprague Dawley rats (male to begin with) singly housed with ad lib access to food and water. Habituated to a food and water restriction schedule of 30 min water and food, then 4.5 hours of food alone, every day for 7 days. On conditioning/perfusion day, rats given 30-min access to 1 of 3 solutions:

  • water
  • 0.05% saccharin + 0.05% Kool-aid (Grape and Cherry counterbalanced)
  • 8% Glucose + 0.05% saccharin + 0.05% Kool-aid (Grape and Cherry counterbalanced).

Rats perfused at different time points after end of access and brains processed for phospho-acetylated histones (need to add specific antibody).

Time Line

  • 2019-June-4 Rats (n=9) arrive
  • 2019-June-5 Remove food and water
  • 2019-June-6 30 min water at 1100-1130, 5 hour food 1100-1600 (Day 1)
  • 2019-June-7 30 min water, 5 hour food (Day 2)
  • 2019-June-8 30 min water, 5 hour food (Day 3)
  • 2019-June-9 30 min water, 5 hour food (Day 4)
  • 2019-June-10 30 min water, 5 hour food (Day 5)
  • 2019-June-11 30 min water, 5 hour food (Day 6)
  • 2019-June-12 30 min water, 5 hour food (Day 7)
  • 2019-June-13 30 min water or kool-aid or kool-aid + glucose (n=3/group); perfuse 1 hour later
